That's usually the hard bit.

I think you just have to play with the settings. Do you have anything
obvious in the mixer controls? Eg "Line In as Center" or "Duplicate
front output" ?
Try plugging some headphones into the line in and see whether you hear
anything?
Also, it's probably easiest to test line in while feeding the signal to
the speakers, rather than recording and playing back.
Lastly, sometimes the buttons take a second or so to have an effect.
